Just curious. I have a Rogers & Spencer with an R&D Conversion Cylinder (6 shot) in 45LC that I have not fired yet. My standard smokeless reload now is 6.2 gr. of TrailBoss and a 200 gr.LRNFP. Will this be a safe load in the R&D. I'll probably stick with BP, but sometimes like top practice and plink with smokeless.

If that meets the typical definiton of a cowboy load, then the cylinder is made to withstand those pressures and probably a bit more.   I have shot all kinds of factory ammo in my various guns with RandD cylinders without problems.  Others will be along shortly to add information I am sure.

I have shot the R&D Cylinders with as much as 8 grains of Unique behind a 250 grain cast RNFP and it was a bit stout for the '58 Remington.  I finally settled for a 165 grain HBRN Bullet ontop of 30 grains of BP and an equivalent volume of APP in a Schofield length .45 Colt case.  (Yeah, I trimmed it back to Schofield length.).  Shot well with that load and with 5 grains of Unique.
Listen!  Do you hear that?  The roar of Cannons and the screams of the dying.  Ahh!  Music to my ears.
